A non-invasive method to quantitate reactive oxygen species in human plasma.
While reactive oxygen species (ROS) have been implicated in numerous pathologies and important regulatory functions in biological systems, definitive detection and quantification of these species in human biological samples has proven to be problematic. This is due, in part, to the extreme chemical reactivity of ROS with critical biomolecules such as proteins, and to the inability of conventional detection methods to differentiate between ROS species. In this paper, we present a new method for detection and quantification of ROS in human plasma, based on the human plasma-derived dipeptidyl peptidase-1 (DPP-1) quenching of the ROS-sensitive dye, DHR. This non-invasive method, uses a catalytic domain-deletion mutant of DPP-1 to directly scavenge ROS produced in a human plasma sample, and was used to provide quantitation of ROS in human plasma without exogenous addition of the ROS sources (e.g., xanthine oxidase/hypoxanthine). This new approach provides a highly-sensitive method to quantitate ROS in human plasma, potentially useful for human disease biomarker discovery.A candlelight vigil was held in front of the Cultural Center of Catalonia (CCC) in Barcelona on Monday evening, marking the 13th anniversary of the still unsolved murder of the Catalan academic and activist in the city.
